A wave start from 7am downtown, finishing at the foot of Campus Martius. The route runs through Woodbridge, the District Detroit, Eastern Market and the Dequindre Cut, and crosses into Canada over the Ambassador Bridge, so a valid travel document is required.

The Detroit Free Press Marathon runs through the streets of Detroit before crossing the Detroit River into Windsor, Ontario, via the Detroit-Windsor Tunnel, then returning over the Ambassador Bridge. The course finishes back in downtown Detroit.
