Loop course on the Longleaf Horse Trails in DeSoto National Forest near Laurel, Mississippi, combining a 12.6-mile and a 6.1-mile loop per distance (the 100-mile runs seven big and two small loops). Mostly singletrack with double-track logging road through rolling pine forest; the longest gap between aid stations is 6.2 miles.

The MS50 is held the first Saturday in March each year on the Longleaf Horse Trails in the DeSoto National Forest just south of Laurel, MS. The race was moved from Leland, MS to its current location in 1996 by Carl Touchstone, a local dentist and accomplished ultra runner.
